How long does equioxx stay in a horses system

“Horses can have a very strong response to this product, so it can be very beneficial,” said Hanson. “Absorption reaches its peak between four to six hours after the first dose. The drug has a fairly long half-life, which means it’s eliminated slowly, so there is some slight accumulation over the first week.

Equioxx, a new NSAID, has a more selective approach.

The first COX-2 inhibitor for treating pain and inflammation in horses has been approved by the Food and Drug Administration and the United States Equestrian Federation.

There are some double-edged swords in the realm of medicine when it comes to keeping our horses comfortable.

Traditional nonsteroidal anti-inflammatory medicines like phenylbutazone (Bute) and flunixin meglamine (Banamine), which are routinely used to treat pain and inflammation associated with osteoarthritis and degenerative joint disease, are a good example. However, long-term or high-dose usage of these medications might result in stomach ulcers, other gastrointestinal issues, and kidney damage.

Merial, a Duluth, Ga.-based animal health firm, developed Equioxx, a novel nonsteroidal anti-inflammatory drug (NSAID) that has the potential to be a helpful tool in the treatment of horse joint discomfort with fewer side effects. Equioxx is the first FDA-approved horse NSAID that targets the cyclooxygenase-2 enzyme while sparing COX-1. It is based on the chemical firocoxib, which has been marketed in chewable tablet form as Previcox for dogs since 2005.

Cyclooxygenase is significant because it converts arachidonic acid present in cell membranes into prostaglandins, which are involved in a variety of physiological functions. COX-1 kicks off a slew of helpful actions, including creating a pleasant mucous lining to shield the stomach from its acidic digesting fluids. COX-2 is implicated in pain and inflammatory potentiation.

Traditional NSAIDs suppress both COX-1 and COX-2, thus they decrease undesirable pain and inflammation while also shutting down the COX-1 pathways’ beneficial effects.

“We’ve been able to put together some very excellent numbers that indicate how medicines like phenylbutazone and flunixin are possibly non-selective, maybe even modestly COX-1 selective,” says the researcher. “You’re inhibiting both enzymes at pretty much any concentration, which is why they work so well, but it’s also one possible explanation for why you see some of the GI, particularly, and some of the renal, side effects,” said Peter Hanson, DVM, PhD, executive director of Merial’s project development group and Equioxx’s research and development project leader.

“We don’t suppress COX-1 at all with firocoxib over the therapeutic range and even at a rather significant dosage,” he noted.

Equioxx was tested in safety tests for 92 days at doses up to 12.5 times the authorized dose of 0.045 mg/lb (0.1 mg/kg) of body weight once daily for up to 14 days. Oral ulcers seemed to be more common at higher dosages, while GI tract ulcers were avoided.

“If you examine horses’ grazing habits, you’ll see little nicks and cuts around their lips. COX-2 is critical in various self-signaling activities for wound healing, and at higher dosage levels, we completely suppress COX-2. “We achieved high levels of inhibition at lesser dosages, but not all day; it would decrease back down to enable the healing processes to take place,” Hanson said.

“What was very intriguing was that we didn’t detect a rise in stomach ulcers or intestinal ulcers in those experiments, even at such high levels where we’re targeting COX-2 100 percent,” he stated. “We did find some alterations in the kidney at extremely high dosages above the therapeutic range for a long duration, much as you see with Bute or flunixin.” Because both COX-1 and COX-2 are active in the kidney, even when we spare COX-1, we still suppress COX-2, so a long-term, high dosage [92 days and 12.5x in the research] might still have an effect.”
