Equioxx was tested in safety tests for 92 days at doses up to 12.5 times the authorized dose of 0.045 mg/lb (0.1 mg/kg) of body weight once daily for up to 14 days. Oral ulcers seemed to be more common at higher dosages, while GI tract ulcers were avoided.

“If you examine horses’ grazing habits, you’ll see little nicks and cuts around their lips. COX-2 is critical in various self-signaling activities for wound healing, and at higher dosage levels, we completely suppress COX-2. “We achieved high levels of inhibition at lesser dosages, but not all day; it would decrease back down to enable the healing processes to take place,” Hanson said.

“What was very intriguing was that we didn’t detect a rise in stomach ulcers or intestinal ulcers in those experiments, even at such high levels where we’re targeting COX-2 100 percent,” he stated. “We did find some alterations in the kidney at extremely high dosages above the therapeutic range for a long duration, much as you see with Bute or flunixin.” Because both COX-1 and COX-2 are active in the kidney, even when we spare COX-1, we still suppress COX-2, so a long-term, high dosage [92 days and 12.5x in the research] might still have an effect.”
